Flow cytometry users already accustomed to an established instrument often do not readily switch to new instruments. When the NYU Langone Health Center Cytometry and Cell Sorting Laboratory acquired two ZE5 Cell Analyzers, the lab addressed this challenge by making use of the new capacity the instruments offered, while ensuring users they would now be able to generate the best data possible. The lab implemented a program designed to demonstrate new instrument capabilities, compare data quality to currently used instruments, encourage participation in comprehensive instrument training, and provide hands-on experience. The ZE5 Cell Analyzers aided this program greatly as the new and improved features, data quality, user experience, and ease of use all contributed to a dramatic rise in instrument use.

The immune system has evolved sophisticated mechanisms to avoid attacking the host body. One such mechanism, T-cell exhaustion, shuts down effector T-cells. Cancer cells exploit T-cell exhaustion to escape being targeted by the immue system. Recently, researchers have used monoclonal antibody drugs to block suppressive cell surface markers to reinvigorate T-cells, restoring their ability to clear cancer cells from the body.
We designed a flow cytometry panel to examine in-depth changes to the T-cell phenotype, exhaustion, and cellular subset identity markers before and after chronic stimulation and in combination with a monoclonal antibody drug treatment targeting the PD1 receptor.
